MOTS-c Overview

MOTS-c (Mitochondrial Open Reading Frame of the Twelve S rRNA type-c) is a 16-amino-acid peptide encoded within the mitochondrial genome. It represents one of the first identified mitochondrial-derived peptides and is studied for its role in cellular metabolic regulation, AMPK pathway activation, and mitochondrial-nuclear signaling communication.

MOTS-c was first identified in 2015 by Lee and colleagues at the University of Southern California, marking a significant discovery in mitochondrial biology. The peptide is encoded by a small open reading frame within the mitochondrial 12S ribosomal RNA gene, challenging the long-held view that mitochondrial DNA encoded only 13 proteins.
